Die Domitilla Katakombe liegt an der alten Via Ardeatina auf dem Grundbesitz der adeligen Römerin Flavia Domitilla, einer Nichte des Flavius Clemens, der im Jahre 95 A.D. Konsul war. Dieser Zweig der Flavier Familie war allem Anschein nach dem Christentum zugetan, denn wir wissen von den Historikern der Zeit, dass Domitian den Flavius Clemens aus religiösen Gründen zum Tode verurteilen ließ und dass seine Frau und seine Nichte auf die Insel Ponza verbannt wurden. Vor ihrem Exil stellte die Nichte des Konsuls ihren Grundbesitz an der Via Ardeatina der christlichen Gemeinde zur Verfügung. Auf diesem Gelände entstand später die größte christliche Katakombe von Rom.
Die wichtigsten Märtyrer der Domitilla Katakombe sind Nereus und Achilleus, zwei Soldaten, die vermutlich Opfer von Diokletians Verfolgung waren (304 A.D.). Sie waren in der dortigen Basilika begraben, einer majestätischen Hallenkirche aus der Zeit des Papstes Siricius (385-399). Die Basilika wird durch Säulen mit wieder benützten Kapitellen in drei Schiffe unterteilt und besitzt eine Apsis (Chorraum) und ein Vestibül (Vorraum). Ein anderer sehr alter Kern ist die unterirdische Grabkammer (Hypogäum) der Flavier. Diese entstand am Ende des zweiten Jahrhunderts als private heidnische Grabkammer. Später aber, während des dritten Jahrhunderts, wurden dort auch Christen begraben, deren Grabstätten mit Szenen aus der Heiligen Schrift verziert wurden. Der Rundgang durch die Katakombe führt auch an der Grabkammer der Veneranda, dem Bogengrab der kleinen Apostel und der Grabkammer des Totengräbers Diogenes vorbei.
