Luftfartsverket uttalte i 1950 at Tromsø, som Nord-Norges største by, burde ha egen flyplass. Stortinget besluttet i 1953 at ny flyplass i nord skulle bygges på Langnes. Først ble det vedtatt å bygge en 1400 meter lang rullebane, men etter noe lobbyvirksomhet av lokale politikere ble det etter hvert bestemt å bygge en 2000 meter lang rullebane for å få ned større fly. Byggingen av flyplassen startet i 1961.

Flyplassen ble åpnet 14. september 1964 av daværende kronprins Harald. Han ankom flyplassen med den første flyvningen kl 11:45 i en Sud Aviation Caravelle fra SAS. Den nye flyplassen i Tromsø la grunnlaget for å etablere flyruter til Finnmark, ettersom SAS-ruter til Finnmark kunne mellomlande i Tromsø. Flyplassen muliggjorde også flyreiser til Longyearbyen.

Det opprinnelige terminalbygget ble erstattet av en ny terminal i 1977, formet som en halvsirkel. Den nåværende terminal ble bygget i 1998, som en del av en omfattende utvidelse og modernisering av flyplassen. I forbindelse med de siste moderniseringene fikk flyplassen et nytt kontrolltårn, en tunnel for å lede nærliggende veitrafikk under en noe forlenget rullebane, og for første gang gangbroer direkte fra terminalbygget til oppstillingsplassene. Enkelte deler av den gamle terminalbygningen er fortsatt i bruk.

Siden åpningen har over 46 millioner passasjerer reist til og fra flyplassen (september 2015).

Civil Aviation Administration stated in 1950 that Tromso as northern Norway's biggest city, should have its own airport. Parliament decided in 1953 that the new airport in the north would be built on Langnes. First it was decided to build a 1400 meters long runway, but after some lobbying of local politicians was eventually determined to build a 2000 meters long runway to get down larger aircrafts. Construction of the airport began in 1961.
The airport was opened on 14th September 1964 by Crown Prince Harald. He arrived at the airport with the first flight at 11:45 a.m. in a Caravelle from SAS. The new airport in Tromsø laid the foundation for establishing routes to Finnmark, since SAS routes to Finnmark transit stop in Tromsø. The airport also made possible flights to Longyearbyen.
The original terminal was replaced by a new terminal in 1977, shaped like a semicircle. The current terminal was built in 1998, as part of a comprehensive expansion and modernization of the airport. Along with the recent modernization a new control tower was built, a tunnel to guide nearby road traffic under an extended runway, and for the first time footbridges directly from the terminal to the pitches. Some parts of the old terminal building is still in use.Since the opening more than 46 million passengers have traveled to and from the airport (September 2015).
