Il newyorkese Larry Daley ha spesso cambiato lavoro e casa, un giorno Larry trova finalmente lavoro al Museo di storia naturale di New York, il cui direttore, spinto da una sempre minore affluenza, ha deciso di assumere un unico custode in sostituzione dei tre vecchi guardiani: Cecil, Gus e Reginald. Costoro forniscono a Larry un elenco di istruzioni da seguire durante la notte, raccomandandogli di non far entrare o uscire nulla o nessuno dal museo.
Quella sera, dopo una breve pennichella, Larry si accorge che lo scheletro del Tyrannosaurus rex si è mosso dal suo piedistallo, per andare a bere alla fontanella. Spaventato, Larry si rende conto che qualsiasi cosa all'interno del museo ha preso vita. Tra i reperti esposti nel museo hanno maggiore risalto il T-Rex ; Dexter, una dispettosa scimmia cappuccino che gli ruba le chiavi; due popolazioni di statuine di antichi romani e di colonialisti del far west , entrambi in competizione per l'espansione del proprio territorio , nella stessa area di un gruppo di statuine di Incas; manichini impagliati di sudisti e nordisti, in guerra tra loro; una scorbutica statua di Cristoforo Colombo; un Moai dell'isola di Pasqua, un gruppo di unni capitanati da Attila e un gruppo di romani Capitanati dal generale ottaviano ; alcune statue di neanderthaliani attratte dal fuoco; la nativa americana Sakagawea, con Lewis e Clark.
Durante le notti successive ne succederanno di ogni nel museo animato da una tavola magica, un furto di opere da parte dei 3 guardiani più anziani e la rivolta per riprendere le opere rubate e la tavola che anima il museo prima che sorga l’alba.
Il mattino seguente, i TG locali parlano di impronte di T-Rex nella neve, pitture rupestri e neanderthaliani sui tetti, pensando che siano trovate pubblicitarie del museo. Il direttore inizialmente licenzia Larry, ma subito dopo cambia idea dopo aver visto la nuova enorme affluenza allo stesso: Larry infatti ha deciso di sfruttare la capacità delle statue di muoversi aprendo il museo anche nelle ore notturne. Ora Nick è fiero di presentare il padre e la sua professione a scuola.
Intanto i tre custodi vengono obbligati da Larry a ripulire tutto il museo dopo le scorribande notturne.
Nel film oltre tanti personaggi primari e secondari un personaggio dice questa citazione

New Yorker Larry Daley has often changed jobs and homes, one day Larry finally finds work at the Museum of Natural History in New York, whose director, driven by an ever decreasing turnout, has decided to hire a single custodian to replace the three old guards: Cecil, Gus and Reginald. They provide Larry with a list of instructions to follow during the night, recommending him not to let anything or anyone enter or exit the museum.
That evening, after a short nap, Larry notices that the skeleton of the Tyrannosaurus rex has moved from its pedestal, to go and drink from the fountain. Frightened, Larry realizes that everything inside the museum has come to life. Among the exhibits exhibited in the museum, the T-Rex stands out; Dexter, a mischievous capuchin monkey who steals his keys; two populations of ancient Roman figurines and colonialists from the far west, both competing for the expansion of their territory, in the same area as a group of Inca figurines; stuffed mannequins of southerners and northerners, at war with each other; a grumpy statue of Christopher Columbus; a Moai from Easter Island, a group of Huns led by Attila; some statues of Neanderthals attracted by fire; the Native American Sakagawea, with Lewis and Clark and the statue of Teddy Roosevelt
During the following nights all sorts of things will happen in the museum animated by a magic board, a theft of works by the 3 oldest guards and the revolt to take back the stolen works and the board that animates the museum before dawn.
The next morning, the local news reports talk about T-Rex footprints in the snow, cave paintings and Neanderthals on the roofs, thinking that they are publicity stunts of the museum. The director initially fires Larry, but soon after changes his mind after seeing the new huge turnout at the same: Larry in fact has decided to exploit the statues' ability to move by opening the museum even at night. Now Nick is proud to introduce his father and his profession at school.
Meanwhile, the three custodians are forced by Larry to clean up the entire museum after their nighttime escapades.
In the movie, besides many primary and secondary characters, one character says this quote
“Every journey, even the longest, always begins with the first step.”
