In the 1980s, a project with the goal of sequencing the human genome started to materialise; or as they stated it: "The ultimate goal of this initiative is to understand the human genome" and "knowledge of the human is as necessary to the continuing progress of medicine and other health sciences as knowledge of human anatomy has been for the present state of medicine.".

The $3-billion project was formally founded in 1990 by the United States Department of Energy and the U.S. National Institutes of Health, and was expected to take 15 years. In addition to the United States, the international consortium primarily comprised geneticists in the United Kingdom, and also in France, Germany, Japan, China, and India.

Due to widespread international cooperation and advances in the field of genomics (especially in sequence analysis), as well as major advances in computing technology, a 'rough draft' of the genome was finished in 2000.

Since then, the technologies for DNA sequencing has advanced at a rapid pace. The most common methods that are used today are often called next-gen sequencing (NGS). In general, these methods sequence a lot of small fragments of the wanted DNA sequence in parallel and it is then attempted to put all these small fragments together. The most common commercial methods are 454 pyrosequencing, Illumina sequencing and SOLiD sequencing.

In the case of SOLiD sequencing, you look on fragments (reads) of 50 bases each. A run of the method on a long DNA sequence will take 1-2 weeks and will generate a total of 30-50 Gigabases of information, i.e. about 60 million short reads will be sequenced. This means that many regions were sequenced multiple times, and will overlap with each other.

All these reads will then have to be put together by computer software in a process called sequence assembly. The software looks for the overlaps between all the short reads and tries to put them together into the real DNA sequence.
