Couffouleux possède une agglomération urbaine récente, datant du XIXe siècle et dont l’origine est liée à la création de la voie ferrée Toulouse-Paris par Capdenac.
Quatre hameaux structurent par ailleurs le territoire communal : Saint-Pierre de Bracou, Sainte-Quitterie, Saint-Victor, Saint-Waast.Chacun de ces hameaux est doté d’une église (Saint Waast et Saint-Pierre de Bracou sont classées monuments historiques).
Couffouleux est l'une des rares communes françaises à être orthographiée de deux manières. Avec deux f selon l'usage local qui bafoue l'étymologie du mot, avec un seul f selon l'usage "administratif"...
